ABSTRACT xii Benign prostatic hyperplasia (BPH) is an age dependent condition that affects old men. The condition is associated with symptoms like frequency in urination, hesistancy, nocturia, weak urine stream and sexual dysfunction. Most BPH patients use conventional methods that include drugs targeting 5-alpha reductase enzyme, the effect of administration of methanol extract of Curcuma longa on liver function, renal profile, body weight and relative organ weight was studied in Benign prostatic hyperplasia (BPH) induced male wister rats. Phytochemical and acute toxicity study was conducted on the Curcuma longa using standard methods. Twenty five animals were divided into five experimental groups of five rats each; Group 1 (Normal control) received olive oil administered orally everyday and by subcutaneous injection of olive oil (every other day) for 28days. Group II (Diseased control) received subcutaneous injection of 0.9mg/kg body weight of dihydrotestosterone/ 0.9ng/kg body weight Estradiol Valarate every other day and oral administration of pure olive oil for 28days. Group III (Drug control) received subcutaneous injection of 9mg/kg body weight of Dihydrotestosterone (DHT)/0.9mg/kg body weight of Estradiol Valarate every other day and oral administration of 5mg/70kg Finasteride every day for 28days. Group IV (lOOmg Curcuma longa) received subcutaneous injection of 9mg/kg body weight of Dihydrotestosterone/0.9mg/kg body weight of Estradiol Valarate every other day and oral administration of lOOmg/kg ofCurcuma longa for 28days. Group V (200mg Curcuma longa) received subcutaneous injection of 9mg/kg body weight of Dihydrotestosterone/0.9mg/kg body weight of Estradiol Valarate every other day for 28days and Curcuma longa 200mg/kg administered orally everyday. Results ofthe phytochemical showed high levels offlavonoid (4.03 ± O.Olmg/lOOg), followed by phytosterol (3.62 ± 0.02mg/100g). Terpene (0.41 ± O.Olmg/lOOg) has the lowest concentration. The result also showed that prostate specific antigen (PSA), which is a maker for BPH decreased significantly (P<0.05) in groups treated with different doses of Curcuma longa extract. There was significant (P<0.05) difference in organ weights, while ALT, AST, Creatinine and urea significant (P<0.05) decrease in treated BPH rats compared to non treated deseased groups. 3 is study reveals important phytochemical constituent which shows that Curcuma longa may have beneficial effect in preventing or suppressing the development of BPH and useful in treatment and management.
