ABSTRACT x This study was carried out to to determine the Salmonella typhi activity and bi ine shrimp lethality value ofCoffee (Coffea arabica') LeafDecoction Formula (Johnu Tisane). The specific objectives of the study included to: determine the antimicrobial activities of Coffee (Coffea aiabica) Leaf Decoction Formula on Salmonella typhi and to determine the brine shrimp lethality value ofCoffee (Coffea arabica) Leaf Decoction Formula. The coffee leaves (Coffea arabica) were processed to obtain John-U tisane decoction. The antimicrobial parameters and lethality analysis were done at Veterinary Laboratory, Michael Okpara University of Agriculture, Umudike; Abia State. The micro-organisms (Salmonella typhi) was clinically isolated from the specimen and were collected in sterile bottles and refrigerated at 0-50C and then subjected to laid down convectional biochemical methods (Gibson and Khoury, 1986; Murray et rzZ.,1985) for identification and preservation. The shrimps eggs stored in refrigerator at 4 °C was hatched with sea water under environmental conditions (temperature, light and humidity) and eggs were poured into sea water (at ratio 1:1000) and it hatched after 24 hours. Actively swimming larva/nauplii were seen in the water. Data were subjected to statistical methods used to analyze the data in this study was Statistical Package for the Social Sciences (SPSS). The results ofthe findings showed inhibitory effects of different concentrations of Coffea arabica leaf decoction on Salmonella typhi. Result revealed antimicrobial activity ofthe extract against gram negative (Salmonella typhi) bacteria, at concentrations of 1ml, 'Ami dilution and 'Ami dilution ofthe extract. However, at 1/g ml, V16 and I/32 ml dilutions ofthe extract, no antimicrobial activity was shown against the gram negative bacteria. Result on the inhibitory effects of different concentrations of Coffea arabica leaf decoction on Salmonella typhi, showed antimicrobial activity of the extract against the gram negative bacteria (Salmonella typhi) at concentrations of 1ml, 'Ami dilution and 'Ami dilution of the extract, while no antimicrobial activity was recorded at Vs"11’ V16 mI and V32 ml dilutions ofthe extract. The present study recorded that the extent of lethality was directly proportional to the concentration ofthe extract. This was observed in the increased rate of death of brine shrimp due to increased concentration ofthe plant extract. After 24 hours of observation, all the shrimp weie suivived in the control. But it was not so with the test groups, as mortalities were observed. The study concluded that plant extracts have great potential as antimicrobial compounds against microorganisms. Thus, they can be used in the treatment of various infectious diseases caused by resistant microbes.
