ABSTRACT xiii Monosodium glutamate, a widely used food additive has been reported to possibly induce oxidative stress. The present study evaluated the in vitro antioxidant potential of Mango seed kernel (MSK) and in vitro protective effect of Mango seed kernel/ethanolic extract (MSKEE) against monosodium glutamate-induced oxidative stress using 20 male albino wistar rats (weighing 80-165g) classified into 5 group (4 rats per group). The rats in group A (control) received portable water and rat chow; rats in treated group B (Negative control) were given 8g/kg/day of MSG orally; rats in treated group C (positive control)) were treated with 300 mg/kg/day of mango seed kernel ethanolic extract orally; rats in treated group D (Low dose extract group) received 200 mg/kg/day Of mango seed kernel ethanolic extract and 8g/kg/day of MSG orally; while rats in treated group E (high dose extract group ) received 8g/kg/day of MSG and 400 mg/kg/day of mango seed kernel ethanolic extract orally. All treatments lasted for 14 successive days. In vitro antioxidant activity of mango seed kernel was measured by DPPH, FRAP and TAC assays. Results indicated that MSG administration resulted in significantly (P<0.05) increased lipid peroxidation (increased malondialdehyde concentration), GPx and GSI1 activity, significantly decreased SOD and CAT activity, and distorted brain morphology in MSG-onlytreated rats in comparison to rats treated with mango seed kernel ethanolic extract. Mango seed kernel treated rats showed preserved tissues compared with distorted tissue in MSG treated rats. Mango Seed Kernel ethanolic extract significantly (P<0.05) reduced lipid peroxidation, improved superoxide dismutase (SOD),catalase and glutathione peroxidase activities, and showed higher free radical scavenging activity in comparison with ascorbic acids. The study concluded that MSKE may offer ameliorative protection against MSG-induced oxidative stress in rats.
