ABSTRACT x Effect of methanolic extract of Annona muricata in biomarkers of oxidative stress in Trypanosoma brucei infected rats were evaluated. Twelve (12) rats were used to test for acute toxicity, the animals were divided into three groups of A, B and C and graded doses of (4000mg/kg, 3000mg/kg, 2000mg/kg) methanolic leaf extract of Annona muricata were administered orally to each rat. They were monitored for signs of acute toxicity such as behavioral changes or death for a period of 24 hours. Thirty albino rats weighing between 73 -250g were used to test for biomarkers of oxidative stress. They were randomly divided into five groups of 6 rats. Group A, served as uninfected untreated, Group B served as infected untreated, Group C was infected and treated diaminazene aceturate at dose of 3.5mg/kg in 5 days post infection, while . Groups D and E were treated orally methanolic extracts of annona muricata at 300mg/kg and 600mg/kg respectively. The mean lipid peroxidation level (MDA) of decreased significantly (P < 0.05) in the uninfected untreated compared to infected untreated and infected treated groups. The malondialdehyde (MDA) was also significantly (P< 0.05) higher in the groups treated with Diaminazene aceturate compare with the group treated with methanolic extract of Annona muricata. The mean superoxide dismutase . (SOD) was significantly (P<0.05) higher in the uninfected untreated group compared to like infected untreated group and is comparable with the groups treated with the extract at 300mg/kg body weigh. The group treated with Diaminazene aceturate was also significantly (P<0.05) lower than the uninfected untreated and is comparable with the group treated with the extract at 600mg/kg. The mean catalase was significantly (P<0.05) lower in the infected untreated group compared to the uninfected untreated group. There - was no significant (P>0.05) different in the groups treated with the extract, at 300mg/kg and 600mg/kg body weight respectively, however the groups treated with Diaminazene Aceturate, was significantly (P<0.05) lower than the groups treated with the extract. In conclusion, methanolic extract of Annona muricata was found to ameliorate the oxidative stress and enhance the antioxidant status of Trypanosoma brucei infected rat. The result of this study indicates that Annona muricata could be used as supplements in trypanosome - endemic area to manage the oxidative stress arising from the disease
