ABSTRACT xiii The effect of administration ofmethanol extract of Urtica dioca leaf on prostate specific antigen (PSA) and antioxidant parameters in dihydrotestosteroneestradiol valarate induced Benign prostatic hyperplasia (BPH) induced male albino rats was investigated. Phytochemical and acute toxicity study was conducted on Urtica dioca using standard methods. Twenty five rats were divided into five (5) experimental groups of five rats each; Group 1-normal control, Group 2-disease control (BPH) group, Group 3-standard drug control (finasteride group), Group 4 and 5 was treated with 100 and 200mg/kg bwt. of Urtica dioca extract respectively. Benign prostatic hyperplasia was induced in all groups except the normal control group for every other day for 28days. Results of phytochemical studies showed high content of alkaloid (4.72 ± 0.02mg/100g) and saponin (4.04 ± 0.03mg/100g). Tannin, flavonoid and phytosterol, were observed at moderate level. Glycoside (1.12 ± O.Olmg/lOOg) and anthocyanide (1.02 ± O.Olmg/lOOg) was observed to be low in the leaf of Urtica dioca. Terpene(0.23 ± 0.02mg/100g) was also observed to be significantly low in the leaf of Urtica dioca. The acute toxicity test was gotten at 3500mg/kg bwt. of the extract. The prostate weight and PSA which is a marker for BPH significantly (p < 0.05) decreased in treated BPH rats compared to disease control group. SOD and GPx activities were significantly (p < 0.05) increased in all the treatment groups when compared with the disease control rats. The MDA level which is a marker for lipid peroxidation was significantly (p > 0.05) increased in disease control when compared with other treatment groups. This study showed that Urtica dioca leaf can serve as a therapy in the management ofBPH which might be as a result ofthe presence phytochemicals contained in the leaf.
