ABSTRACT Sphenocentrum jollyanum is a major consumed plant in Nigeria indigenous system of folklore medicine claims that root part of the plant is used in the treatment ofsome disease such as diabetes mellitus, malaria, cough, stroke, and inflammatory condition. The anti-hyperglycaemic effect of the aqueous root extract of Sphenocentrum jollyanum was evaluated in normal and in alloxan induced diabetic rats. The level of glucose was determined at different doses 200mg/kg and 400mg/kg and time treatment with plant extract and glibenclamide. Then the rats were fasted for 12hours and then sacrificed, Blood sample were collected for further determination of enzymatic antioxidant level such as Catalase (CAT) ,Superoxide dismutase (SOD), Glutathione (GSH) and malondialdehyde (MDA) in alloxan induced diabetic rats. The result shows for CAT that high dose 400mg/kg had (2.46±0.01) followed by low dose 200mg/kg (2.06±0.01) and lastly control group had (3.77±0.07) respectively. The result of SOD showed that high dose (400mg/kg) had (0.83±0.01) followed by low dose (200mg/kg) had (1.20±0.02) and lastly, control group had (0.88±0.01) respectively, MDA showed that high dose 400mg/kg had (4.72±0.01) followed by low dose 200mg/kg had (4.63±0.01) and lastly control had (4.63±0.01) respectively the result showed for GSH that high dose 400mg/kg had (4.36±0.02) followed by low dose (200mg/kg) had (4.59±0.06) and lastly control group had (7.95±0.09) respectively. The result suggested that CAT, MDA, and GSH with high dose 400mg/kg showed significant (P<0.05) increase than low dose 200mg/kg and control, were as the result suggested that SOD with high dose 400mg/kg is non-significant (P>0.05) higher than low dose 200mg/kg and control, the result suggested that oral administration of aqueous root extract of Sphenocentrum Jollyanum to diabetic rats showed significant (P<0.05) reduction in hyperglycaemia and increases antioxidant action in alloxan induced diabetic rats.
