Antimicrobial Activity Of Grape Vitis Vinjfera L. Against Food Borne Pathogens:- Mgbamgba Blessing I

ABSTRACT / ix This study determined the antimicrobial activity of vitis vinifera L. seed extracts against food borne pathogens. The Grape seed was obtained from local market in Umuahia, Abia State, Nigeria were prepared by dissolving 0.2g (200mg) of the plant extract in 2ml of (DMSO) Dimethyl Sulfoxide to give 200mg/ml stock solution. The strains of Microorganism used as test were Staphylococcus aureus, Escherichia coli, Klebsiella spp. Bacillus spp. Aspergillus niger and Aspergillus flavus. This study revealed that the In-vitro antimicrobial activity of the methanol and ethanol soluble extracts of the Grape seed varied against the test isolates. The highest in-vitro antimicrobial activity (17.0mm) was exhibited by the Grape (Vitis vinifera L.) seed ethanol extract at the highest concentration of 200mg/ml against Klebsiella spp. while the least antimicrobial activity (6.0mm) was exhibited at the concentration of 25mg/ml and !2.5mg/ml against all the test organisms. The methanol extract had highest in-vitro antimicrobial activity of (18.0mm) at a concentration of 200mg/mI against Escherichia coli, while the least antimicrobial activity (6.0mm) was exhibited at the concentration of 25mg/ml and 12.5mg/ml against all the test organisms except the fungal test organisms which was exhibited at the concentration of 50mg/ml respectively. It was also revealed in this study that the methanol extract of Grape (Vitis vinifera L.) seed was more effective as shown in the diameter zones of inhibition when compared to the ethanol seed extracts of Grape (Vitis vinifera L.). The minimum bactericidal concentration revealed that the methanol and ethanol soluble extracts of Grape (Vitis vinifera L.)« was active against all the test bacteria, but showed greater activity against Escherichia coli, Klebsiella spp, Aspergillus niger and Aspergillus niger at 200mg/ml. it can be concluded that both methanol and ethanol extracts of Grape (Vitis vinifera L.) have broadspectrum antimicrobial activity when compared to the standard antibiotics used in this study, hence can serve as natural therapeutic agent against some food borne pathogens.
