ABSTRACT

This study showed the effect of methanol extract of Malay apple leaf on PSA and lipid profile parameters in dihydrotestosterone - estradiol valarate induced Benign Prostate Hyperplasia in male Wister rats. Thirty (30) male Wistar rats aged between 10 - 12 weeks, divided into five groups of 6 rats each were used in this study. The lethal dose (LD50) is the concentration at which half of the population dies was also determined. The lethal dose (LD50) of Syzygium malaccense was found to be 5000mg/Kgbwt. Benign Prostate Hyperplasia was induced by subcutaneous injection of 9mg/kg dihydrotestosterone (DHT) and 0.9mg/kg estradiol valerate dissolved in olive oil every other day for 28 days. The groups received different treatments, group 1- normal control, group 2 -disease control, group 3- drug control, group 4 - lOOmg Malay apple leaf extract, and group 5 -200mg Malay apple leaf extract. Biochemical parameters were assayed using standard procedures. Result showed no significant difference (P > 0.05) between relative prostate weight of group 3 (Drug control) and group 5 (Rats administered with 200mg/kg bwt of extract). No significant difference was observed in group 4 rat when compared to normal control rats. The relative prostate weight of group 2 rats increased significantly (P < 0.05) compared to all other groups. There were no significant difference (P > 0.05) in the relative heart, liver and kidney. Serum prostate specific antigen increased significantly in group 2, 4 and 5 compared to group 1 and 3. Cholesterol level decreased significantly (P< 0.05) in group 5 (2.29mg/dl ± 0.22mg/dl) while HDL - cholesterol increased (1.75mg/dl + 0.13mg/dl) relative to group 3 (1.85mg/dl + 0.05mg/dl). In group 2 cholesterol and LDL - cholesterol increased significantly (P > 0.05) (7.04mg/dl ± 0.12mg/dl and 4.73mg/dl + 0.16mg/dl while HDL - cholesterol decreased significantly in (P < 0.05) (0.29mg/dl ± 0.05mg/dl). This finding suggests that Malay apple leaf contain bioactive agents that may be useful in the management of Benign Prostate Hyperplasia and its complications, and can be of pharmacological importance in the management ofBenign Prostate Hyperplasia.
