ABSTRACT

I Monosodium Glutamate (MSG), which is chemically known as AJI-NO-MOTO also familiar as MSG in routine life. MSG is always considered to be a controversial food additive used in the world. This work was aimed at investigating the therapeutic dose effects of artemether_lumefantrine on kidney histology and some antioxidant indicators in the kidney homogenate of normal and monosodium glutamate_intoxicated rats. Adult male Wistar rats (n = 5) were six(6) groups each were treated as follows: Group I (control); group II: Therapeutic dose of artemether_lurnefantrine or Artemisinin-based combination therapy(ACT); group III: Overdose of ACT; group IV:A known toxicant (MSG at 8000mg/kg/w);group V:Therapeutic dose + MSG at 8000mg/kg/w; group VI: Overdose + MSG at 8000mg/kglw. Administration was orally and twice daily by accurately weighing 4 tablets of the antimalaria drug that an average 70kg man takes twice daily and dissolving it in lOOmI of distilled water and administered in volume corresponding to the rats weight while the MSG was done once, for a period 7 days. At the end of the experiment, animals were sacrificed and the kidney was taken and Superoxide dismutase (SOD) activity and zinc concentration were determined. There was a significant (p<O.05) decrease in the SOD activity and zinc concentration of the MSG-only rats, when compared with the control rats which improved in the MSG + therapeutic dose of ACT. It can be concluded that the therapeutic dose of artemether_lumefantrine elicited a significant in vitro antioxidant property and may protect against MSG-induced oxidative stress by increasing SOD activity and zinc concentration.
