ABSTRACT
The
liver plays a pivotal role in the metabolism oftoxic substances that enter the
body. Thioacetamide is a toxic substance which induces hepatotoxicity from its
metabolic conversion to free radicals products: thioacetamide sulfoxide and
thioacetamide- Soxide which attacks microsomal lipids leading to their
peroxidation and production of reactive oxygen species. 1 his study was
conducted to investigate the hepatoprotective effects of the methanolic extract
of the rind of Citrullus lanatus on Thioacetamide (TAA) induced oxidative
stress in male albino wistar rats. The study was conducted using live groups of
five animals each. Group A served as the Normal control group which received
distilled water and grower’s mash only. Group B served as the Negative control
and received TAA (300mg/kg intraperitoneally once). Group C (Positive control
group) received Silymarin (lOOmg/kg body weight of rats only). Group D and E
rats received TAA (300mg/kg intraperitoneally once) and were administered
methanolic rind extract of Citrullus lanatus (250mg/kg and 500mg/kg body weight
respectively) for seven days. Animals were subsequently anaesthetized with
chloroform, and blood samples collected for liver and renal function assays.
There were significant increases (P< 0.05) in the serum levels of ALT, AST,
ALP, Bil and MDA in the negative control group compared to the normal control,
positive control and test groups. There was also a significant decrease (P<
0.05) in the level of TP, CAT and SOD activity in the negative control group
compared to the normal control, positive control and the test groups. The
results from the animal groups treated with the extract revealed that the
methanolic extract of Citrullus lanatus may have hepatoprotective effects,
which may have ameliorated the damage caused in the liver by TAA induced
toxicity
