ABSTRACT

The present study investigated the glutathione peroxidase activity in the serum and liver homogenate of normal and monosodium glutamate-intoxicated rats co-treated with artemeter and lumafantrine. 30 male wistar albino rats weighing 80-140g were randomly sectioned into 6 groups(n=5), as follows: Normal group: feed and water only; ALC therapeutic dose group: 0.17ml ALC, feed, and water; ALC overdose group: ALC therapeutic dose multiplied by 5(0.8ml), teed and water; MSG-only group: MSG (8mg/g bodyweight), feed and water; MSG+ALC therapeutic dose group: ALC therapeutic dose(0.17ml) + MSG (8mg/g bodyweight), feed and water;MSG+ALC (overdose): ALC (overdose) (0.85) + MSG (8mg/g bodyweight), feed and water. After 7-days of treatment the rats were humanely sacrificed and blood samples and liver tissue collected for glutathione peroxidase assay. Results showed less than normal liver glutathione peroxidase activity in all treatments in the order: ALC (overdose) (19.72±1.59)> ALC (therapeutic dose) (17.51±1.56)> MSG+ALC (therapeutic dose) (15.77±0.55)> MSG+ALC (overdose) (15.56±0.70)> MSG-only rats (13.32+1.21). Mean serum glutathione peroxidase activity of MSG-only rats (1.67+0.13) was significantly (P<0.05) lower than Normal (4.60+0.36), ALC (therapeutic dose) (3.82+0.38), ALC (overdose) (4.62+0.22), MSG+ALC (therapeutic dose) (3.66+0.37), and MSG+ALC (overdose) (7.50+0.33) groups respectively. Liver homogenates showed higher Gpx activities than serum. The study confirmed a possible deleterious effect of MSG and ALC administration on glutathione peroxidase activity in serum and liver homogenates as evidenced by reduced Gpx activities especially in liver ofMSG-treated rats, however further studies will help explain the paradoxical increase in serum Gpx activity seen in ALC overdose treated rats.
