ABSTRACT
The
effect ofthe administration of methanol extract of Curcumin longa on Prostate
specific antigen level, relative organ weight and antioxidant parameters was
studied in dihydrotestosterone — estradiol valarate induced benign prostatic
hyperplasia (BPH) in male Albino Rats. Phytochemical and acute toxicity study
was conducted on the Curcuma longa using standard methods. Twenty rats were
divided into five experimental groups of 4 rats each; Group 1- normal control,
Group 2- disease control (BPH-group), Group 3- standard drugs control
(finasteride-group), Group 4 and 5 (100 and 200 mg/kg of Curcuma longa extract
respectively). The normal was not induced with BPH but was given olive oil
administered orally (everyday) and subcutaneously (alternative days) for 28
days. The disease control group was induced BPH hormones in an alternative days
and oral administration of olive oil lor 28 days. The standard control group
(induced BPH) was given finasteride 5mg/70kg administered orally every day. The
lOOmg/kg and 200mg/kg of methanol extract of Curcuma longa was given to BPH
induced rats orally every day respectively. Results of the phytochemical
studies showed high content of flavonoid (4.030 ± 0.014mg/100g) followed by
alkaloid and tannin (2.405±0.007 and 2.225 ± 0.021mg/100g respectively).
Terpene showed the lowest concentration (0.405 ± 0.007mg/100g). The acute
toxicity tests showed that the methanol extract of Curcuma longa is not toxic
up to 4500mg/kg. The result also showed that prostate specific antigen (PSA)
which is a maker of BPH decreased significantly (P < 0.05) in groups treated
with different doses of Curcuma longa extract. Superoxide dismutase and glutathione
peroxidase activity, was observed to be significantly (P < 0.05) decreased
in disease control group rats compared to the treated groups while a
significant (P < 0.05) increase in Malondialdehyde level was observed in the
disease control group animals compared to normal control and treated groups.
This study reveals important phytochemical constituent which shows that Curcuma
longa may have pharmacological efficacy in preventing or suppressing the
development ofBPH and useful in its treatment and management.
