ABSTRACT
This study evaluated the
potentials of Bacillus and Aspergi/lus to produce amylase enzyme. Soil sample
were collected from different sites, diluted serially and plated on Tryptone
Soya agar and Sabouraud Dextrose agar. The Bacillus Specie were identified
using biochemical characterization protocol and screen for amylase production
on nutrient agar amended with 1% starch. Among the Bacillus species isolated,
four (4) isolates exhibited amylolytic power with hydrolysis halo diameter of
between 12mm to 18 mm. Maximum amylase productions was observed for the
Bacillus megaterium species with its diameter zone of clearance of 18.0mm on
starch agar. In a similar way, one of the Aspergillus niger species isolated in
this study produced clear haloes. The amylolytic ratio from this study showed
that on three Bacillus isolates produced amylolytic ratio greater than 3
(good), and only one (1) showed less than 2 (poor). It could be concluded that
this study has demonstrated the suitability of exploitation of the soil habitat
for isolation of Bacteria and fungi capable of amylase production which can be
used for several industrial applications
